Used at non-normal angles
of incidence, these longpass and shortpass filters highly reflect one
specified spectral region while optimally transmitting another. In
fluoresence microscopy the excitation light is reflected onto the
sample by the dichroic, usually at an angle of incidence of 45°.
The emitted fluoresence light is transmitted through the dichroic and
onto the detector. It is crucial that the dichroic has good
reflection and transmission characteristics and that the transition
is a sharp as possible.
